Effects of BCG-polysaccharide nucleic acid and OVA allergen combined with chitosan nano sustained-release vaccine on inflammatory cells and cytokines in bronchoalveolar lavage fluid of asthmatic mice

[Abstract] Objective To explore the effects of BCG-polysaccharide nucleic acid (BCG-PSN) and ovalbumin (OVA) allergen combined with chitosan (CS) nano sustained-release vaccine on inflammatory cells and inflammatory factors in bronchoalveolar lavage fluid (BALF) of asthmatic mice. Methods A total of 80 mice were randomly divided into 8 groups (group A was normal control group, group B was asthma group, group C was blank CS group, group D was CS-BCG-PSN group, group E was CS-OVA group, group F was BCG-PSN group, group G was CS-BCG-PSN-OVA vaccine group, group H was hormone group). Except for the normal control group and asthma group, asthmatic mice in each group received intraperitoneal injection of corresponding drugs on the 18th and 24th day.The mice in the hormone group were injected with dexamethasone intraperitoneally before atomization-induced asthma.The asthma group and the normal control group were given the corresponding dose of normal saline at each time point. 48 hours after the last induction,the related cytokines (IL-4 and IFN-γ) and the count of leukocytes and eosinophils in BALF were measured. Results The number of leukocytes and eosinophils and IL-4 in group D, group E, group F and group G were lower than those in group B and group C. The decrease of leukocyte,eosinophil count and IL-4 in asthmatic mice treated with CS was more significant than that in BCG-PSN and OVA alone(P<0.05).Moreover,the levels of IFN-γ in group D, E, F and G was higher than that in asthma group and blank CS group, and the increase of IFN-γ in asthmatic mice treated with BCG-PSN combined with OVA was higher than that in BCG-PSN and OVA separately(P<0.05). Conclusion CS-BCG-PSN-OVA complex can regulate the number of cells and the level of cytokines, which may be used to correct the imbalance of Th1/Th2 proportion and further alleviate airway inflammation,thereby providing a theoretical basis for the treatment of asthma.
