藏药灰枝紫菀生药学鉴别

【摘 要】 目的:研究藏药灰枝紫菀的性状及显微特征,为其生药学鉴别提供理论依据。方法:采用性状、显微鉴别方法研究藏药灰枝紫菀的生药学特性。结果:灰枝紫菀茎多分枝,帚状,皮灰褐色,撕裂,嫩枝纤细,被密短糙毛或柔毛。叶上表面被短糙毛,下表面被柔毛。头状花序在枝端密集成伞房状或单生;花序梗细,长1~2.5cm,有疏生的苞叶。灰枝紫菀根皮层宽广,可见众多石细胞,皮层内侧可见4~6个分泌道。茎横切面呈圆形,外被角质层,木质部导管呈放射状排列。叶上、下表皮均有腺毛,厚角组织1~2层,栅栏组织紧密分布,气孔不定式,3~4个保卫细胞。粉末黄棕色。腺毛和非腺毛较多。冠毛为多裂性分枝状,各分枝为单细胞,花粉粒黄色,具3孔沟,表面具尖刺状雕纹,石细胞单个散在,呈类方形,层纹及孔沟较明显,无色。可见花冠裂片表面具棒槌状突起。导管多为网状纹理和螺纹导管。偶见草酸钙簇晶和方晶。结论:灰枝紫菀干燥药材性状特征,根、莖、叶横切面和叶表面显微特征,以及全草粉末显微特征均可显示其独特鲜明的生药学特性,以性状特征和全草粉末特征最为突出,为灰枝紫菀的生药学鉴定和质量标准制定提供参考。

Abstract:Objective To research on characters and microscopic features of Aster poliothamnus Diels, for its basis of pharmacognosy identification. Methods The pharmacognosy characterist-ics of Aster poliothamnus were studied by means of the characteristic and microscopic features. Results There were many ramifications on the stem of Aster poliothamnus, broom shape,gray-brown bark,tear,shoot tenuity and covered with density short-rough hair or villose.The upper sur- face of the leaves was covered with short-rough hair and the lower was villose.The capitulum gl-omerated in corymbiform on the terminal of the branches or solitary,the peduncle was slender,1~ 2.5cm long and the bracts were sparse.The root cortex of Aster poliothamnus was wide and there were numerous sclereids and 4~6 secretory canals could be seen on the inside of cortex.The tra-nsverse section of stem was round,covered with cuticles and vessels in xylem were arranged rad-ially.The glandular hair existed in the upper and lower epidermis,one to two layers of collenchy-ma.The palisade tissues were distributed densely,the stomas were infinitive,3~4 guard cells,yell-ow brown powder,many glandular and non-glandular hairs.Pappus was multicolumn branched,e-ach branch was single cell,the pollen was yellow, 3-colporate, echinate sculpture on the surface, sclereids?scattered individually,showed quasi-square,the laminated striations and colporate were obvious,colourless.Wooden club-shaped protuberance could be seen on the surface of the corolla lobe. The vessels were mostly netlike textures and spiral vessels.Calcium oxalate crystals and c-ubic crystals could be observed occasionally. Conclusions The character and feature of dried A-ster poliothamnus,the microscopic features of the transverse section of the root,stem,leaf and leaf surface as well the whole herb powder all showed unique and distinct pharmacognostic properti-es.The character and feature and the features of the whole herb powder were the most prominen-t.This study could provide scientific references for pharmacognostic identification and the form-ulation of quality standardization.

菊科植物灰枝紫菀Aster poliothamnus Diels干燥花序是藏医传统习用药材“露琼”的基原药材之一,藏医药经典著作《晶珠本草》记载“可解毒,治温病时疫、木保病,清脉热”,可用于流行性感冒发烧、食物中毒等[1]。汉民族民间多用其全草治疗流行性感冒和发烧,有关灰枝紫菀的研究较少,亦未见生药学研究报道。本论文通过系统观察灰枝紫菀全草生药学特征,为藏药“露琼”基原药材之一—灰枝紫菀的生药学鉴定和质量控制提供参考。
1 仪器与材料
1.1 仪器 XY-200型高速多功能粉碎机,GZX-9070MBE电热鼓风干燥箱。日本Nikon H550S显微镜,Nikon DS-Ri2显微成像系统,TS100(Nikon) 倒置生物显微镜,德国徕卡Leica RM2245轮转式切片机,TKY-BMB型石蜡包埋机。
1.2 试剂与材料 甘油,苯酚,水合氯醛,明胶,乙醇,甲醛,冰醋酸,二甲苯,番红,固绿,加拿大树胶,进口石蜡均为分析纯。
灰枝紫菀药材采自青海省久治县德合龙寺,经江西中医药大学钟国跃研究员鉴定为菊科Compositae紫菀属Aster灰枝紫菀Aster poliothamnus Diels 的干燥花序。标本现存于江西中医药大学中药资源与民族药研究中心标本馆。
2 方法
2.1 性状描述 结合相关文献和标本对灰枝紫菀药材进行性状描述。
2.2 显微鉴定 取灰枝紫菀药材2份,1份干燥后粉碎,过5号筛,用于药材粉末鉴定;1份置水中煮沸软化,分拣出根、茎、叶,制成石蜡切片,进行显微观察。另取适量经水软化展平的叶片,用解剖针或刀片撕取上下表面,制成临时制片用于叶表面显微观察。
3 结果
3.1 药材性状 灰枝紫菀茎多分枝,帚状,皮灰褐色,撕裂,嫩枝纤细,叶密集,被密短糙毛或柔毛。叶多脱落或破碎,完整叶片展开为多长圆形或线状长圆形,长1~3cm,宽0.2~0.8cm,全缘,基部稍狭或急尖,边缘平或稍反卷;上表面被短糙毛,下表面被柔毛,两面有腺点。头状花序在枝端密集成伞房状或单生;花序梗细,长1~2.5cm,密被绒毛。总苞圆盘形,中央管状花。子房棕色,密被长冠毛。气香,味淡。
3.2 显微鉴别
3.2.1 根横切面 截面为圆形,表皮细胞排列紧密,多皱缩或脱落,木栓细胞5~10列,栓内层明显,最内层细胞外壁增厚。皮层宽广,有细胞间隙,有分泌腔和草酸钙棱晶。中柱鞘纤维束散在,次生韧皮部明显。木质部一直分化到中心,射线宽1~2列,导管单个散在,木纤维与木薄壁细胞区别不明显。如图1所示。3.2.2 茎横切面 木栓层细胞数列,内含红棕色物。皮层和韧皮部均不明显。木质部占大部分,射线宽1~3列细胞,导管壁较薄,单个散在或数个切向连接,略呈放射状分布,木纤维与木薄壁细胞区分不明显。中央髓部细胞多有空洞或裂隙,外圈木质部常伴有纤维束,形成层呈环分布,如图2所示。
3.2.3 叶横切面 上、下表皮细胞各1列,排列紧密,多呈类方形,可见非腺毛,多由2~4个细胞组成, 均被角质层。上下表皮内均可见栅栏组织,紧密分布。中脉维管束外韧形,下表皮内有厚角组织2~3列,皮层薄壁组织大,内含草酸钙结晶,导管呈放射状排列。如图3所示。
3.2.4 叶横切面 上表皮表观细胞呈多边形或类方形,可见腺鳞,垂周壁较平直,呈扁球形,由6~7个细胞排列在同一平面上。下表皮细胞表观呈波纹不规则形,可见腺鳞和非腺毛,气孔不定式。如图4、图5所示。
3.2.5 灰枝紫菀粉末 本品粉末黄棕色。腺毛大多棒槌状,头部稍膨大,和腺柄均多由2个细胞组成。非腺毛较多,大型,多弯曲,顶端钝圆,微木化,14个细胞组成,直径可达10μm,较粗者壁有网状纹理。厚壁细胞呈长方形,非木化,纹孔较多,排列成纵行。冠毛分枝状,各分枝为单细胞,先端渐尖。花粉粒常见,具3孔沟,表面具尖刺状雕纹。石细胞单个散在,呈类方形,层纹及孔沟较明显,无色。可见花冠裂片表面具棒槌狀突起。导管多为网状纹理和螺纹导管。偶见草酸钙簇晶和方晶。如图6所示。
4 讨论
藏药“露琼”的基原药材之一是菊科植物灰枝紫菀A. poliothamnus的干燥花序[2],故《卫生部药品标准藏药分册》收载了其粉末显微鉴别特征。考虑到汉族地区民间多用其全草治疗流行性感冒,发热等[3],而有关其全草的生药学研究鲜有报道。本文对灰枝紫菀干燥全草的性状和显微特征进行研究,确认其均可反映灰枝紫菀药材独特的生物学特性和组织特征,其中药材的性状特征和全草粉末显微特征尤能反映其生药学特性。此研究结果可作为灰枝紫菀药材生药学鉴别的参考依据。
