吲哚7位的修饰研究

摘要:吲哚又名苯并吡咯,是常見的六元并五元氮杂环,具有抗癌、消炎、杀菌等活性,在香料、农药、食品等领域有广泛应用。本文研究了该衍生物7位的修饰,生成的产物结构被晶胞图证实。

吲哚衍生物可用作消炎药、Puallone CDK抑制剂、DY-9760e钙调蛋白拮抗剂,是应用最广泛的杂环衍生物之一(图1)。国内外诸多课题组都报道了关于该类骨架的构建。从已有合成方法中吸取经验,探究更加高效地合成该类衍生物的方法,值得进一步思考。
一、合成方法
在乙酸酐溶液中,烯胺酮和苊醌反应生成了7位羟基化的吲哚。在乙醇溶液中,烯胺酮和苊醌反应生成了7位醚化的吲哚。两个反应的温度均为120℃(图2)。
二、晶胞图
培养了该类衍生物的单晶,晶胞图如图3、图4、图5所示,证实了结构的准确。
三、结语
在120℃的乙酸酐或乙醇溶液中,烯胺酮和苊醌发生串联反应,构建了吲哚骨架,并分别实现了该骨架7位的羟基化和醚化。晶胞图证实了产物结构的准确。
