Abstracts: The effects of Vitamin B1 on the growth and development and physiological and biochemical indexes of Dioscorea bulbifera L. plantlets were studied. The results showed that when vitamin B1 concentration was 0.25 mg/L, the number of bud, the bud length, the root number, the root length, the leaf number, the leaf length and the plant height could increase the number of Dioscorea bulbifera L. plantlets. When the concentration of vitamin B1 increased, the morphological indexes of Dioscorea bulbifera showed a decreasing trend. The results of physiological and biochemical indexes determination showed that when vitamin B1 concentration was 0.25 mg/L, the total chlorophyll content and the activity of POD and SOD increased, but the contents of malonaldehyde(MAD) and proline, the membrane permeability decreased. When the concentration of vitamin B1 increased, the total chlorophyll content and the activity of POD and SOD decreased, while the contents of MAD and proline, the membrane permeability increased. Under the conditions of containing vitamin B1, the content of proline of plantlets was significantly positively correlatied with the content of MDA and themembrane permeability, but there was significant negative correlation between the total content of chlorophyll and the activity of POD and SOD. The changes of content of proline can be used as a physiological index to determine the growth and development inhibition degree of D. bulbifera L. plantlets..

黄独(Dioscorea bulbifera L.)为薯蓣科薯蓣属植物,是世界广布种,在中国主要分布于浙江、江西、福建等地[1]。

1.1 材料

黄独试管苗,由上饶师范学院生命科学学院植物组织培养室提供。主要使用仪器有三角瓶、高压灭菌锅,超净工作台等。

1.2 方法

将黄独带芽茎段(0.5~1.5 cm)接种至100 mL三角瓶中进行单因子试验。每个三角瓶均加入液体培养基(MS+KT 2.0 mg/L+NAA 0.1 mg/L)40 mL。试验处理分成5组,各组的维生素B1浓度依次为0、0.25、0.50、1.00、2.00 mg/L。以上培养基pH调节为5.8~6.0,配制后在高压灭菌锅中灭菌20 min(121 ℃、0.1 MPa),接种都在超净工作台上进行。接种后的培养条件如下:光照时间14 h/d,光照度1 000~1 600 lx,温度25 ℃,湿度70%~80%。每隔7 d观察并记录黄独带芽茎段新生芽数、根数、叶片数、芽长、根长、叶片长和株高。培养第50天对5组试管苗的生理生化指标进行测定,总叶绿素(叶绿素a和叶绿素b)含量的测定采用丙酮比色法[6],脯氨酸含量的测定采用酸性茚三酮比色法[6],过氧化物酶(POD)活性的测定采用愈创木酚法[6],超氧化物歧化酶(SOD)活性的测定采用NBT光还原法[6],丙二醛(MDA)含量的测定采用硫代巴比妥酸法[6],细胞膜透性的测定采用相对电导率法[6]。试验重复3次,所有统计数据均表示为平均值±标准差。各组试验数据用SPSS 19.0统计软件进行One-Way ANOVA分析,再进行LSD法检验。
